antimicrobial peptide brevinin-2r induces the secretion of a pro-inflammatory cytokine in hepg2 cells
Authors
abstract
introduction: antimicrobial peptides as the body's defense strategy play an important role in resistance against infection of microorganism. these peptides are able to modulate the immune and inflammatory processes through the production of defensive molecules. therefore, the modulatory effects of brevinin-2r, an antimicrobial peptide extracted from the skin of the frog (rana ridibunda), was evaluated in this study. materials and methods: mtt assay was conducted for evaluating the cytotoxic effect of brevinin-2r on human liver carcinoma cells (hepg2). then, real-time pcr was used to assess the inflammatory properties of this peptide. results: it was appeared that the brevinin-2r has a low toxicity against hepg2 cancer cells and can reduce cell growth to 24%. results of molecular analysis showed that brevinin-2r increases the expression of il-1b and il-6 genes in cancer cells in a dose dependent manner. conclusion: the results show that brevinin-2r has a regulatory role in inflammation through targeting the genes involved in the process.

full textCationic antimicrobial peptide, magainin down-regulates secretion of pro-inflammatory cytokines by early placental cytotrophoblasts
BACKGROUND Human placental villous cytotrophoblasts exhibit relative externalization of negatively charged moieties to the outer leaflet of the plasma membrane during the time of syncytialization rendering their reactivity to positively charged cationic antimicrobial peptides (CAMPs) during the window of implantation and early placentation. Vaginal administration of a synthetic CAMP, Ala(8,13,1...
full textNaegleria fowleri Lysate Induces Strong Cytopathic Effects and Pro-inflammatory Cytokine Release in Rat Microglial Cells
Naegleria fowleri, a ubiquitous free-living ameba, causes fatal primary amebic meningoencephalitis in humans. N. fowleri trophozoites are known to induce cytopathic changes upon contact with microglial cells, including necrotic and apoptotic cell death and pro-inflammatory cytokine release. In this study, we treated rat microglial cells with amebic lysate to probe contact-independent mechanisms...

Background and Objective: Peptide as an agent in manufacturing new drugs has recently been taken into consideration. Brevinin-2R is a nano peptide with a wide range of biological actions. Thus, the current study was designed in order to evaluate the in vitro effects of Brevinin-2R on oral squamous cell carcinoma cells in comparison to normal mice fibroblast cells. Materials and Methods: In t...
full textIn Vitro Cytotoxic Activity of a Lactococcus lactis Antimicrobial Peptide Against Breast Cancer Cells
Background: Nisin, an effective natural food preservative, is an antimicrobial peptide produced by Lactococcus lactis. Although it has been mainly studied and developed as a potential alternative for antibiotics, other pharmacological effects of the nisin including cytotoxic and anti-tumor activity have been attracted many attentions. Objectives: Here, we ...
full textNiacin Modulates Pro-inflammatory Cytokine Secretion. A Potential Mechanism Involved in its Anti-atherosclerotic Effect
The pathogenesis of atherosclerosis includes the assignment of a critical role to cells of the monocyte/macrophage lineage and to pro-inflammatory cytokines. Niacin is known to improve lipid metabolism and to produce beneficial modification of cardiovascular risk factors. The aim of this work was to investigate if Niacin is able to modulate pro-inflammatory cytokine production in macrophages in...
